Operation steps of bile bacteriolysis test:
1 test tube method
1) For the test tube, scrape / pick out the suspicious colony to be tested with rayon swab (do not use medical cotton swab) or inoculation ring into the small test tube of bile salt solution, elute, squeeze the swab head against the wall to emulsify it to form a relatively thick bacterial suspension (about 1~2# Michaelis standard turbidity concentration), squeeze the excess liquid of the swab head against the tube wall, and screw on the test tube plug;
For negative control, take another small test tube of normal saline (with the same volume as bile salt solution), adjust it into bacterial suspension with the same turbidity by the same method, and screw on the test tube plug as the negative control; If necessary, Streptococcus pneumoniae can be used as a positive control;
2) 35 ℃± 2 ℃, incubation for 5min ~ 15min; Compared with the negative control tube, observe whether the bacterial solution in the small test tube becomes clear or the turbidity is significantly reduced. See the figure below.

Colony dissolution method: dip 1-2 full inoculation ring bile bacteriolytic reagent, drop it on a single suspicious typical colony to be tested, cover it, and incubate at 35-37 ℃ for several minutes, no more than 30 minutes. The Streptococcus pneumoniae colony will dissolve and disappear, leaving only grass green marks in its place. Streptococcus viridis is not dissolved.
